Medical Lab Science Prerequisite Requirements

To qualify for ASCP MLS certification under Route 2, candidates must complete a bachelor's degree plus specific biology and chemistry coursework. While exact requirements vary by program, the ASCP standard requires:

Biology

16 semester hours, including one semester of microbiology

Chemistry

16 semester hours, including organic chemistry or biochemistry

Mathematics

College-level math (often pre-calculus or higher)

Statistics

One semester for data analysis competency

Anatomy & Physiology

Often required by bridge and master's programs

Clinical Experience

Documented clinical lab training (arranged separately)

Note: Survey or "non-majors" science courses generally do not count toward the requirement. Many programs also require science prerequisites to be completed within the last 5–10 years.

ASCP Certification Pathways

The ASCP Board of Certification (BOC) is the credentialing body for clinical lab professionals. To sit for the MLS exam, candidates must qualify under one of several routes. Most career changers and MLT bridge candidates qualify under Route 2 or Route 4.

What's the difference between a Medical Laboratory Scientist (MLS) and a Medical Laboratory Technician (MLT)?
Medical Laboratory Scientists typically hold a bachelor's degree, perform more complex testing, validate methods, troubleshoot unusual results, and often take on supervisory or quality-management roles. Medical Laboratory Technicians typically hold an associate's degree and work under the supervision of an MLS, performing routine testing. The two credentials map to different ASCP BOC exams (MLS vs. MLT).
How many biology and chemistry credits do I really need for ASCP MLS Route 2?
16 semester hours of biology, including at least one semester of microbiology, plus 16 semester hours of chemistry, including at least one semester of organic chemistry or biochemistry. Survey or non-majors courses generally don't count toward the requirement. Always verify current rules at the ASCP BOC website before enrolling.
Can I take these prerequisites if my science degree is more than 5 or 10 years old?

Many MLS, pathologists' assistant, and histotechnology programs require science prerequisites completed within the last 5 to 10 years. Will my target NAACLS-accredited program accept online prerequisites?
Most NAACLS-accredited programs accept prerequisite coursework from regionally accredited institutions. What happened to ASCP MLS Route 3?
Route 3, which previously served Clinical Laboratory Assistant (CLA) candidates, was discontinued effective January 1, 2023. Candidates who would previously have qualified under Route 3 now generally apply under Route 2 (bachelor's + coursework + clinical experience) or Route 4 (MLT bridge), depending on their existing credentials.
Do I need clinical experience to sit for the MLS exam?
Yes. Under most ASCP routes (including Route 2), candidates need a documented period of qualifying clinical laboratory experience or training, typically completed at a hospital, reference lab, or NAACLS-recognized clinical site. How long does each course take?
